Product Description
Dofetilide/Dofetilidum is a class III antiarrhythmic agent. It is marketed under the trade name Tikosyn by Pfizer, and is available in the United States in capsules containing 125, 250, and 500 µg of dofetilide. It is not available in Europe or Australia.
Due to the pro-arrhythmic potential of dofetilide, it is only available by prescription from physicians who have undergone specific training in the risks of treatment with dofetilide. In addition, it is only available by mail order or through specially trained local pharmacies.
Production method
N-methyl-4-nitrophenethylamine and 2-[4-nitrophenoxy] ethyl bromide were dissolved in acetonitrile, potassium carbonate and sodium iodide were added, and the reflux reaction was stirred. Concentrate until dry, and the remaining oily solids are reprocessed to obtain the compound. The resulting compounds were dissolved in ethanol and catalyzed by Raney nickel. Strain and concentrate the filtrate until dry. The treated compounds and methanesulfonic anhydride are dissolved in dichloromethane and stirred at room temperature. The product is further treated to give dofetilide.
Use
Dofetilide/Dofetilidum is a specific class III antiarrhythmic drug and a selective potassium channel agonist. It can treat and prevent atrial arrhythmias, such as atrial fibrillation, atrial flutter and paroxysmal supraventricular tachycardia, and also prevent the occurrence of ventricular tachycardia. This product is a selective potassium channel agonist and a class III antiarrhythmic drug. It can prolong the action potential duration (equivalent to QIC interval) and effective refractory period of ventricular myocytes, but has no significant effect on heart rate, blood pressure, PR interval or QRS wave time, and has no effect on conduction and sinus node function. Mainly used in the treatment of atrial flutter, atrial fibrillation, ventricular tachycardia, ventricular fibrillation and other tachyarrhythmias, the curative effect of up to 80%~100%.
